  • What are the main claims of the paper and how important are they?

    This article  presents  the comprehensive protocols and steps evaluating different antioxidant defence parameters and oxidative stress parameters of rat testis.

    Besides, methods for isolation of different testicular subcellular fractions, germ cell isolation and testis histological preparation were also described. Ready to use protocols containing a lot of information for studying the process of oxidative stress and ROS accumulation in the testis tissure. The article is very useful scientifically.

    The article contains ready to use protocols with detailed steps. The original methods were also cited along with the modiications which would be helpful for the readers for having a knowledge about the original protocols.

    This paper is not a novel work , not useful for others for seminars or for search. Author must feel compatible why he selected rat testis for  such a important work.

    This article describes methods for isolation of different testicular subcellular fractions, germ cell isolation and testis histological preparation, in addition to describing steps evaluating different antioxidants and oxidative stress parameters of rat testis. Although the assays themselves are very important in redox-related research, it is unclear how important they are with respect to rat testis.

    Yes, as it pertains to the protocols that were referenced and used. No, from this paper alone, one could not reproduce anything. There is not nearly enough detail describing experiments.

    Yes. Author's methods apart from reference methods should be elaborated. For example, description of mitochondrial isolation and purification was reduced to apparently one centrifugation step in an unknown medium. Typically, mitochondrial isolation is done by Percoll density centrifugation, among other methods. Also, should include a reason for using rat testis and why they are a good analytical system/model. Author should emphasize what they did that was novel/distinct from methods already published on the literature. Description of sperm comes out of no where. How was it extracted? “Mitochondrial fraction was freeze thawed three times” – why? Freeze/thawing at all is not necessarily a good thing; so why did this occur? Germ cell isolation has little to no description. What is the purpose for the tissue, sperm, and cells. Why do all three systems? What can each of these systems tell you that is different from the other?

    Though a review of currently published protocol is useful, this paper falls very short. What is the purpose of rat testis and these particular assays of redox? Grammar and sentence structure is poor. Manuscripts should include an introduction, results, and discussion, in combination with the methods.
